US Route 52 begins at the Canadian border in a town called Portal in North Dakota. US Route 52 becomes Highway 39 as it crosses into Canada. On its way southeast it crosses Minnesota, Wisconsin, Illinois, Indiana, Ohio, West Virginia, Virginia and North Carolina to its end at Number 2 Meeting Street and White Point Gardens along the Charleston Harbor.

US Route 52 crosses from Illinois into Indiana about 5.5 miles west of Kentland, Indiana. On its southeastern journey through Indiana it crosses Newton, Benton, Tippecanoe, Clinton, Boone, Hendricks, Marion, Hancock, Shelby, Rush, Franklin, and Dearborn Counties where it hitches a ride with Interstate 74 at Exit 169 on its way into Ohio.

US Route 52 crosses into Dearborn County from Franklin County about 2.7 miles from its intersection with Interstate 74 at Exit 169. Exit 169 serves as the exit for West Harrison, Indiana. At this point it merges with Interstate 74 and continues east in conjunction with the interstate.  It exits Dearborn County and Indiana about 2.25 miles east as it passes into Ohio.

In Dearborn County US 52 passes through no towns or cities in Dearborn County.
