Interstate 65 runs north south, beginning at an intersection with Interstate 10 near Mobile, Alabama and ending at an intersection with U.S. Route 12 and U.S. Route 20 just southeast of Gary, Indiana. On its way north Interstate 65 crosses Clark, Scott, Jackson, Bartholomew, Shelby, Johnson, Marion, Hendricks, Boone, Clinton, Tippecanoe, White, Jasper, Newton, and Lake counties.

Interstate 65 is enters Clark County from Louisville, Kentucky on the Dr. Martin Luther King Bridge which crosses the Ohio River. Interstate 65 crosses Jeffersonville, then Clarksville . It intersects the Interstate 265 loop about 8.5 miles north at Exit 6A and 6B. It bisects Sellersburg with access to the town at exits 7 at Indiana State Road 60 and 9 at Indiana State Road 311. US 31 runs parallel to Interstate 65 along this route with access points at Exits . About 16 miles north of this intersection Interstate 65 intersects Indiana State Road 160 at Henryville, Exit 16. About 4 miles north of Interstate 65 exits Clark County into Scott County.

It is approximately 4.5 miles to the next exit, Exit 41, at its intersection with Indiana State Road 250. 8.5 miles north of this Interstate 65 intersects US Route 50 just east of Seymour, Indiana at Exit 50. From this intersection, Interstate 65 veers northwest about 5.5 miles to an intersection and interchange with Indiana State Road 11 at exit 55. Interstate 65 leaves Jackson County and enters Bartholomew County about 1.25 miles northwest.
